Are there colors that will not go transparent with .gif

i creating .gif file 1 color transparent. dark yellow. reason, not go transparent when saving web. when select , click transparent, color go transparent. have fideled of .gif settings.

a gif image has 256 color palette. when save image
saving web-safe? or adaptive?

if former, , color have selected not web-safe color,
e.g., #336699, closest web-safe color 1
selected.
